Colocalization of oxytocin and phosphorylated form of elongation factor 2 in the rat hypothalamus.

چکیده
Oxytocin (OT) is one of the neuropituitary hormones and is synthesized in the neurons of the paraventricular nucleus (PVN) and supraoptic nucleus (SON). Previous studies have shown that the mRNAs encoding OT are delivered from the soma to both dendrites and axons of the neurons in the PVN and SON. However, it has not been elucidated whether a translational regulation mechanism to enable local synthesis of the hormone exists in the axons of the neurons of PVN and SON. Elongation factor 2 (EF2) is essential for polypeptide synthesis during protein translation. Moreover, phosphorylation of EF2 by EF2 kinase enhances the translation of certain mRNA species. In the present study, in order to shed light on the mechanisms involved in the translational regulation of OT synthesis, we investigated the localization of phosphorylated EF2. Phospho-EF2 was localized in the soma of the neurons in PVN and SON, and in the swellings of the median eminence where axonal tracts of the neurons in the PVN and SON exist. The phosphorylated form was also observed in the rat hypophysis. Moreover, phospho-EF2 and OT were colocalized in a part of the neurons in the PVN and SON. These results suggest that OT may be partially translated in the axons of neurons in the PVN and SON, and then secreted from the pituitary.

Background: Comparison of the effect of lactation on expression of RFRP-3 mRNA in dorsomedial hypothalamic nucleus (DMH) and KiSS-1 mRNA in arcuate nucleus (ARC) of hypothalamus in lactating and nonlactating rat Materials and Methods: Fourteen rats of the Sprague- Dawley strain were randomly allotted into three groups. The non-lactating rats (n=5) were separated from their pups immediately upon...
متن کاملInhibitory effects of oxytocin on the inflammatory parameters and vascular endothelial growth factor (VEGF) in the rat air pouch model of inflammation
Introduction: The aim of the present study was to evaluate the effect of oxytocin on the angiogenesis and inflammatory parameters in air pouch model of inflammation. Methods: Inflammation was induced by injection of carrageenan into pouches in male Wistar rats. Oxytocin (4.25, 8.5 and 17 μg) was administered intra pouch at the same time as the carrageenan and then for 2 consecutive days. Af...
متن کاملLactation effect on the mRNAs expression of RFRP-3 and KiSS-1 in dorsomedial and arcuate nuclei of the rat hypothalamus
Introduction: Arginine-phenylalanine-amide-related peptide-3 (RFRP-3) and kisspeptin are believed to be inhibitor and stimulator of gonadotropin releasing hormone (GnRH) secretion, respectively. The aim of the present study was to compare the effect of lactation on the expression of RFRP-3 mRNA in dorsomedial hypothalamic nucleus (DMH) and KiSS-1 mRNA in arcuate nucleus (ARC) of hypothalamus...
متن کاملP-222: Expression of RFamide-Related Peptide-3 and KiSS-1 mRNA in The Hypothalamus of Pregnant Rat
Background: There is no follicular development during pregnancy in the rat comparable to the changes of the estrous cycle. RFamide-related peptide-3 (RFRP-3) and kisspeptin were recognized as regulator of gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH) secretion in several species, including the rat. The aim of the present study was to evaluate the relative expression of RFRP...
متن کاملStress activation of IL-6 neurons in the hypothalamus.
An emerging literature attests to the ability of psychological stress to alter the inflammatory cytokine environment of the body. While the ability of stress to cause cytokine release is well established, the neural pathways involved in this control have yet to be identified. This study tests the hypothesis that IL-6 neurons of the hypothalamo-neurohypophyseal system (HNS), a neural pathway pro...
